- Liquid chromatography (LC) with UV 205nm was developed to monitor sweetener “stevioside, aspartame, sucralose and M.S.G” in soju which was made in Korea. The method uses a single-step dilution for sample preparation.
The LC separation was performed on a YMC-pack C18 (2.1 mm x 150 mm) with a mobile phase of acetonitrile and water (35: 65).
The method was validated. The validation parameters included range (10-100ppm), precision, linearity, and accuracy. Standard solutions at 4 concentration levels were prepared for the validation work, including the tests for precision, linearity, and accuracy.
The solutions were prepared in triplicate for each concentration. The relative standard deviation for the precision test was <3% for all 4 concentration levels. The correlation coefficient for the linearity within the concentration range was determined to be > 0.999. The average recovery ranged from 95.7 to 101.1% for the soju samples spiked with these sweetener standards. The detection limit for sweetener was estimated at 0.1~10 ppm.
The method was used to screen 11 kinds of soju samples; stevioside was detected range from 6.2 to 25.2 ppm and the other sweeteners were detected below the detection limit.
