- In recent years, dietary supplements, herbal medicines and counterfeit medications containing the phosphodiesterase-5 inhibitor
( PDE5-I) for erectile dysfunction (ED) are increasing all over the world. It is well known that analysis of the contents of the
adulterated PDE5-Is shows inconsistent doses of active pharmaceutical ingredients and alterna
tive or un-labeled ingredients.
Therefore, it is very important to both identify and quantify the active ingredient. In this study, the liquid chromatography-high
resolution mass spectrometry (LTQ/Orbitrap) method has been applied for the simultaneous determination and identification of
nine most commonly adulterated PDE5-Is including sildenafil, tadarafil, vardenafil and their structural analogues (homosildenafil,
hongdenafil, hydroxy homosildenafil, udenafil, pseudovardenafil and amino tadalafil). The method has been successfully applied
to the analysis of practical samples of seven adulterated supplements and medications.
