Abstract
Glutamate is the predominant excitatory neurotransmitter in the brain and activates two major receptor classes: the ionotropic glutamate(iGluR) and the metabotropic glutamate receptors (mGluR). mGluRs typically play a regulatory role on fast glutamatergic effects by modulating the ion channel activity and/or neurotransmitter release. And there are eight subtypes of mGluR (mGluR 1-8). Among them, mGluR5's over stimulation has been implicated in numerous CNS disorders.2-methyl-6-(phenylethynyl)pyridine (MPEP) is well known potent and selective noncompetitive mGluR5 antagonist. So we, in an attempt to develop novel mGluR5 antagonists as potential therapeutic agents for neuropathic pain, prepared a series of MPEP derivatives.
