- The cells have many binding moieties on their surface for bio-receptors like aptamers. This might result that different aptamers isolated by SELEX using whole cells can bind to different outer parts of cells. In this study, based-on this, we examined the binding assay of various combinations of four different DNA aptamers which were newly isolated against E. coli in our lab. As a result, two among four aptamers showed the competitive binding. We assumed that this is due to similar binding moiety of two aptamers on the surface of E. coli. In addition, the aptamer cocktails composed with mixture of different DNA aptamers showed a higher fluorescence signal by binding of aptamers to E. coli compared with using only single aptamer. This suggested that the usage of aptamer cocktails can enhance the sensitivity in the detection of whole cells. To demonstrate this, the electrochemical detection system was applied. As a result, the sensitivity of E. coli detection was enhanced one-order of magnitude compared with usage of single aptamers.
