LC-Ag+CIS/MS/MS and LC-ESI/MS/MS for analysis of anabolic androgenic steroids

Anabolic androgenic steroids (AAS) are synthetic derivatives of the male hormone, and widely used to improve the athletic performance in sports. The analysis of AAS is generally performed by gas chromatography-mass spectrometry (GC-MS). More recently, liquid chromatography-tandem mass spectrometry (LC-MS/MS) is replacing classical GC-MS methods. However, LC-MS/MS methods for detection of AAS deal with a limited number of AAS. It is related to difficulties of ionization in atmospheric pressure interfaces such as electrospray ionization (ESI) due to the absence of acidic or basic moieties in their structure. In this study, we investigated the ionization and analysis of AAS based on liquid chromatography metal ion coordination ionspray mass spectrometry (LC-M+CIS/MS/MS). Metal ion coordination ionspray mass spectrometry (M+CIS-MS) is a powerful tool in the ionization of non-polar substances such as AAS. Charged complexes ([M+Ag]+) were formed via the addition of silver ions and the AAS were successfully ionized and detected through Ag+CIS/MS/MS. The present LC-Ag+CIS/MS/MS method was compared with LC-ESI/MS/MS method for analysis of 65 exogenous AAS. Only 45 exogenous AAS were detected by LC-ESI/MS/MS method. On the other hand, all AAS including the steroids known as poor ionization in ESI were determined by LC-Ag+CIS-MS/MS method. In addition, the sensitivity of LC-Ag +CIS/MS/MS method is better than LC-ESI/MS/MS method.
