Abstract
Ethnopharmacological relevance: The modified-Chungsimyeolda-tang(DG)isanimportanttraditional Koreanherbalformulausedintraditionalorientalmedicinefortreatmentofcerebrovasculardisorders, including stroke.Theformulaisbasedonthebook “Dongui SasangShinpyun”. Aim ofthestudy: In thepreviousstudies,theneuroprotectiveeffectofDGisdemonstratedinan in vitro Parkinson'sdisease(PD)model,andinthisstudy,the1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) animalmodelofPDisusedtoevaluatethebehavioraleffectofDGandpossiblemechanism through anti-apoptosisofDG.6-Hydroxydopamine(6-OHDA)alsoisusedtoevaluatetheanti-apoptosis effect ofDGinSH-SY5Ycells. Materials andmethods: MPTP wasusedtoevaluatethebehavioraldamageandneurotoxicityinmice.The bradykinesia symptomwasmeasuredbyaPoletestandaRota-rodtestinmice.Alsothelossoftyrosine hydroxylase(TH)-positiveneuronsinducedbyMPTPwasexaminedbyanimmunohistochemicalassay. The DG-mediatedanti-apoptosiseffectwasmeasuredusinganimmunoblottingassaywithapoptosis- relatedmarkerssuchasBaxandcleavedcaspase-3.DGand1-methyl-4-phenylpyridinium(MPPþ) were co-treatedwithprimarydopaminergicneuronstoevaluatetheprotectiveeffectofDG.Theexpressionof caspase-3 andPARPwasmeasuredtodetecttheprotectiveeffectofDGfromthedamageby6-OHDA. Results andconclusions: The treatmentwithDGresultedinprophylacticeffectsonMPTP-inducedPar- kinsonian bradykinesiaandtheimmunohistochemicalanalysisshowedthatDGprovidedtheneuro- protectionagainsttheMPPþ-induced dopaminergicneuronslossthroughtheanti-apoptosiseffect.The present resultssuggestedthatitmightbepossibletouseDGforthepreventionofsubstantianigrapars compacta (SNpc)degenerationinducedbyexposuretothetoxicsubstances,suchasMPTP/MPPþ, inPD mouse model.
