Abstract
Cancerstemcells(CSCs),sidepopulationcontainedfrombulkatumor,aremaincontributorsthathavetheabilitytocancerchemo-resistance,invasion,metastasis,andrecurrence.Phenetyhlisothiocyanate(PEITC)isoneofglucosinolatederivedfromcruciferousvegetables,whichisknownaschemopreventivereagentsinvariouscancercelllines.Inthisstudy,weinvestigatedPEITCreducesNanogtranscriptionalactivityandalsoreducestheexpressionofpluripotencymarkersinhumanembryoniccarcinomacellline.Then,weshowedthatPEITCinhibitsthepropertiesofCSCsincludingclonogenicity,sphereformation,andexpressionofCSCsmarkersOct4,Nanog,Sox2,andALDH1inHCT116stem-likecells.Moreover,toevaluatethepreventioneffectsofPEITC,weestablishedtheCSCsxenograftpreventionmodelusingbalb/cnudemice.Ourresultsshowedthatpre-treatmentofPEITCsuppressedtheexpressionofpluripotencymarkersandprocaspase3andcleavedcaspase9,andparpapoptoticproteinthanPEITCnormaltreatmentandoxaliplatincombinationgroup.Furthermore,weexamineddifferentialgeneexpressionineachgroupbyRNA-seqanalysis.Thus,PEITCmaycontributetopreventingordelayingcolorectalcancergrowthbyinhibitingCSCsviaregulatingtheseveralmolecularfunctionalpathways.
