- Ginseng was traditionally and widely used for Asian folk medicine. In this study, we evaluated the structural changes of ginsenosides in microwave irradiated ginseng extract and demonstrated the anticancer effect of this extract in SK-MEL-2 human melanoma cell line. After microwave irradiation, several kinds of ginsenosides, such as Rg1, Rb1, Rb2, Rc, Rd, Re, were decreased, while other kinds of ginsenosides, like Rg3, Rg5, Rk1, were increased. Especially, the concentration of Rk1 and Rg5 was dramatically increase in this extract. We treated the microwave irradiated ginseng extract to melanoma cell line, cancer cell death was remarkably increased in microwave irradiated ginseng extract treatment. The result of anti-proliferation effect showed an IC50 value of 217.3 mg/mL. Also we tested the ginsenoside fraction from the microwave irradiated ginseng extract to melanoma cell line, obtained an IC50 value of 64.8 mg/mL. It indicates that cytotoxic effect of microwave irradiated ginseng extract came from the increment if specific ginsenosides. The cell death of melanoma cell line was caused by apoptotic pathway. Therefore, microwave irradiation is helpful method to induce the anti-melanoma effect of ginseng extract by enhance the concentration ginsenosides, Rg3, Rg5 and Rk1
